Description
We conducted transcriptome analysis and proteome analysis to discover new genes that may be involved in regulating mitochondrial function. A high-fat diet (60% fat) was administered for 2, 8, and 16 weeks, and RNA seqeuncing analysis was performed on mouse brown adipose tissue to select genes that may be involved in regulating mitochondrial function. box. At the same time, we isolated mitochondria from mouse brown adipose tissue under the same conditions, analyzed the mitochondrial proteome through LC-MS/MS analysis, and discovered commonly selected genes and proteins in both the transcriptome and the proteome.

Sample processing protocol
After cell pellets were carefully washed in PBS buffer, cell pellets were lysed by 5% SDS. And performed by sonication at a setting of 20% energy and on/off cycle 3sec/3sec each for a total of 60 seconds. The lysis sample was heating 95°C for 10min and centrifuged at 12,000 g for 10 min. The supernatant was transferred to a new tube. Protein concentration was then measured using the BCA protein assay (BCA Protein Assay Kit, Pierce).
