cells were grown to A600 of ~1 at 30 Degree C in 3ml of SC(-Leu) medium. Each culture (1 ml of cell suspension at A600 of 1) was washed twice in 1ml of distilled water, serially diluted 5-fold in 200 ml of distilled water, and then 5 ml of each cell suspension was spotted onto SC(-Ura) or SGly(-Ura) plates containing 2 percent fermentable glucose or 2 percent respirable glycerol, respectively. These plates were then incubated at 30 Degree C for 2 or 4 days. |
