This study determined the cryo-EM structure of the active complex of Olfr412, a native mammalian class II odorant receptor. Olfr412 is the mouse ortholog of human OR1D2, and the active complex consists of Olfr412, the odorant methyl-trans-cinnamate, the Gs protein, and Nb35. The odorant-binding pocket of Olfr412 is located deeper within the transmembrane domain than that of the previously reported class I odorant receptor OR51E2. This pocket is composed of poorly conserved hydrophobic residues, providing a molecular basis for broad odorant recognition in class II ORs. Structural analysis and molecular dynamics simulations further revealed that the conserved Y6x55 plays a key role in odorant recognition and activation, functionally paralleling R6x59 in class I ORs, while its interaction with the conserved ECL2 residue E45x51 further stabilizes Y6x55. These findings uncover structural mechanisms underlying odorant recognition and activation in class II ORs.

이 연구는 자연형 포유류 2형 후각 수용체인 Olfr412의 활성 복합체 구조를 Cryo-EM 기법으로 규명하였다. Olfr412는 인간 OR1D2의 쥐 상동 수용체이며, 본 연구에서 규명한 활성 복합체는 Olfr412, 후각 물질 methyl-trans-cinnamate, Gs 단백질 및 Nb35로 구성된다. Olfr412의 후각 물질 결합 주머니는 기존에 보고된 1형 후각 수용체 OR51E2보다 막관통 영역 내 더 깊은 위치에 형성되어 있다. 이 결합 주머니는 서열 보존성이 낮은 소수성 잔기들로 구성되어 있으며, 이는 2형 후각 수용체가 다양한 후각 물질을 인식할 수 있는 분자적 기반을 제시한다. 구조 분석과 분자동역학 시뮬레이션을 통해, 1형 후각 수용체에서 보존된 막관통 나선 6 상단부의 아르기닌에 대응하는 위치의 티로신이 2형 후각 수용체의 후각 물질 인식과 활성화에 핵심적인 역할을 함을 밝혔다. 이 티로신은 세포외 루프 2의 글루타민산과 상호작용하여 수용체의 활성 상태를 안정화한다. 이를 통해 본 연구는 2형 후각 수용체의 후각 물질 인식과 활성화에 관여하는 구조적 기전을 밝혔다.
