This project aims to elucidate the higher-order clustering architecture of BAFF–BAFFR and BAFFR–TRAF3 signaling complexes, which regulate B-cell survival and immune responses in lipid membrane environments. Through cryo-electron microscopy (cryo-EM) analysis, we determined the high-resolution three-dimensional oligomeric structure formed by the ligand–receptor–adapter assembly on the membrane. This study provides fundamental molecular insights into receptor activation and downstream signal transduction, contributing to the development of targeted therapeutics for autoimmune diseases.
프로젝트의 국문 설명
본 프로젝트는 지질막 환경에서 B세포 생존 및 면역 반응을 조절하는 BAFF–BAFFR와 BAFFR-TRAF3 신호전달 복합체의 고차원 클러스터링 구조 규명을 목표로 합니다. Cryo-EM(초저온 전자현미경) 분석을 통해 세포막 상에서 리간드-수용체-어댑터 단백질이 형성하는 고해상도 삼차원 올리고머 구조를 규명하였습니다. 본 연구는 수용체 활성화 및 신호 전달의 분자적 메커니즘을 제시하여 자가면역질환 표적 치료제 개발에 기여합니다.
