This study identified a human-derived peptide sequence capable of forming complexes with mRNA for intracellular delivery. The human beta-defensin 23 peptide sequence forms complexes with various model mRNAs—including EGFP, firefly luciferase, and ovalbumin mRNA—and effectively delivers them into cells to facilitate the expression of the encoded proteins. It is anticipated that this peptide sequence can be utilized to complement existing LNP-based mRNA delivery systems.

이 연구는 mRNA와 복합체를 형성하여 세포 내로 mRNA 전달할 수 있는 인간 유래 펩타이드 서열을 발굴하였다. 인간에서 유래하는 human beta-defensin 23 펩타이드 서열은 모델 mRNA인 EGFP mRNA, firefly luciferase mRNA, ovalbumin mRNA 등과 복합체를 형성하여 세포 내로 mRNA를 효과적으로 전달함으로써 mRNA가 코딩하는 단백질이 발현될 수 있도록 한다. 이러한 펩타이드 서열은 기존 LNP 기반 mRNA 전달체와 상호 보완적으로 활용될 수 있을 것으로 기대된다.

HEK293 cells (2.5 × 104) seeded on a confocal dish, treated with FAM-labeled mRNA or hBD23/mRNA complexes (N/P = 20) were incubated at 37 °C for 4 h. After washing two times with PBS, cells were fixed with 4% formaldehyde solution for 10 min, and the nucleus was stained with Hoest34580.
