Library Construction
Single cell sample preparation for constructing libraries and sequencing was processed in the use of the Chromium controller according to the 10x Single Cell 3??v2 protocol 10x Genomics. To acquire 10,000 targeted cells, the cells were diluted in nuclease-free water and mixed with master mix. Thereafter, the single cell suspension was loaded with Single Cell 3??Gel Beads and Partitioning Oil GEM into a Single Cell 3??Chip. Following to generate the GEMs, single cells were uniquely barcoded and subsequently cDNAs were used to construct libraries according to the manufacturer's instructions. The libraries were sequenced using HiSeq 2500 Illumina
