Chromatin tracing of the Pparg locus in differentiating C3H10T1/2 cells overexpressing TAZ

This study provides chromatin tracing data of the Pparg locus to investigate how TAZ overexpression alters 3D genome organization during adipogenesis. Using Optical Reconstruction of Chromatin Architecture (ORCA) via sequential FISH, we mapped the chromatin structure surrounding the Pparg gene in C3H10T1/2 cells at Day 0 and Day 1 of differentiation. Our findings demonstrate that TAZ overexpression significantly disrupts the enhancer-promoter interactions that are typically established during adipocyte lineage specification, providing a structural basis for TAZ-mediated inhibition of differentiation.
