1. Establishment and validation of a severe allergic asthma mouse model 2. Immune cell isolation from the lungs in severe asthma mice and single-cell multiomic (transcriptome-epigenome) analysis 3. Identification of immune cell types and their subpopulations related to severe allergic asthma induction and drug (steroid) responsiveness through single-cell transcriptome analysis 4. Investigation of molecular mechanisms including signaling pathway-epigenome-gene expression related to severe asthma induction/drug responsiveness through integrated analysis of transcriptome and epigenome data for these cell subpopulations 5.
